ABSTRACT

Objective To investigate the dynamic changes of articular cartilage destruction and nitric oxide nitric oxide/inducible nitric oxide synthase (NO/iNOS).Methods Forty healthy adult male Wistar rats were randomly divided into control group (free movement) and experimental group (overtraining).Total knee joints of rat were collected at week 0,1,2,3,and 6,fixed with 4% PFA,embedded in paraffin,sectioned and stained.Cartilage morphological changes were observed by Safranin-O staining,NO/iNOS content changes in serum and synovial fluid were determined by ELISA,Mankin's scores were obtained,and cartilage pathological changes were analyzed.Results Overtraining resulted in cell sorting disorders,loss of matrix,destruction like rough surface and cracks,and degeneration of cartilage pathology process.Levels of NO/iNOS in serum and synovial fluid increased after overtraining:week 1 (48.088 ± 1.534),(3.058 ± 0.073);week 2 (54.574 ± 0.800),(6.630 ± 0.524);week 3 (68.020 ± 3.184),(7.384 ± 0.306);weeks 6 (104.394 ± 8.094),(11.364 ± 0.882) Levels of NO/iNOS in synovial fluid group were:week 1 (65.814 ±1.610),(8.126 ±0.724);week 2 (72.766 ± 2.344),(9.694 ± 0.564);week 3 (88.816 ± 0.819),(11.326 ± 1.473);week 6 (120.204 ± 12.044),(15.166 ± 1.198),and high level of concentration was found with the increasing amount of exercise.Conclusion There is a positive correlation between the levels of NO/iNOS in synovial fluid or serum and the extent of the damage of articular cartilage.

ABSTRACT

Objective To observe how exogenous estrogen influences the distribution and the expression of NOS positive neurons in the supraoptic nuclei of hypothalamus in the overiectomized female rats. Methods The 2-3-month-old female rats(n=40) were selected as the healthy and nulli-copulatory experimental animals. Rats were divided into following groups: normal control group(n=10), ovariectomized control group(n=10), and two experimental groups that have been injected with estrogen for post-operative 40 days(n=10) and for post-operative 70 days(n=10). Finally, all the animals were infused and the brains were removed. Immunohistochemical (SABC) method was adopted to count the number of NOS poitive neurons and observed the NOS poitive neuronal morphology under the light microscope. The image analysis system was used to test the average gray value of immunoreactivity in NOS positive neurons. Results In the ovariectomized control group, the density of NOS positive neurons in supraoptic nucleus was significantly increased and their shapes were bigger than that of the normal control group(P<0.05). The density and the form of the NOS positive neurons in supraoptic nucleons had no apparent difference between the estrogen for post-operative 40 days group and the ovariectomizeed control group(P>0.05).In the group after estrogen-injection 2 months compared with the normal control group, and the ovariectomized control group, both of the NOS positive neurons' density and the size become significantly decreased, and the staining of cells was lesser in the group injected with estrogen for post-operation 70 days. Conclusion The present results suggest that exogenous estrogen may influence the distribution and the expression of NOS positive neurons in supraoptic nucleus of hypothalamus of ovariectomized female rats.

ABSTRACT

Objective To observe whether or not long-term inhalation of nitrous oxide can affect the cardiac function in rats. Methods Sprague-Dawley rats of either sex were randomly divided into four groups. Group A: inhaled common air for 50 days as control; Group B: inhaled 500mL/L N_2O for 15 days, two hours every day; Group C: inhaled 500mL/L N_2O for 30 days, two hours every day; Group D: inhaled 500mL/L N_2O for 50 days, two hours every day. Float glass intracellular microelectrode recording technique was used for observation of the duration (MAPD_(90), MAPD_(50) and MAPD_(20)) and amplitude of action potentials (APs) of left ventricular muscle cells in vitro. Angiotensin-2 (Ang-2) and eNOS were detected by immunohistochemical technique. Results ① Compared with that in Group A, the Aps duration of left ventricular muscle cells (MAPD) in Group B had no significant change while the MAPDs in Group C and Group D were extended significantly. There were no significant differences among the four groups in Aps amplitude. ② The expression of Ang-2 did not differ significantly between Group B and Group A. The expression level was higher in Group C and D than in Group A. ③ The expression level of eNOS was significantly lower in Group C and D than in Group A (P<0.05). Conclusion Long-term inhalation of N2O can significantly affect the cardiac function in rats, which may be related to higher expression of AngⅡin the heart induced by the long-time excitation of sympathetic nerves.

ABSTRACT

Objective To construct new model of experimental abdominal aortic aneurysm (AAA) and detect the AAA outer diameter and the change of induce nitric oxide synthase(iNOS) in the abdominal aor-tic aneurysm in different period,and analysis the correlation between experimental AAA and iNOS.Methods During the operation of the experiment group,the vascular prosthesis of PTFE was implanted to the rabbit abdominal aorta to form an aneurysm,while sham operation was done in the control group.The tissue of ab-dominal aortic aneurysm was harvested in 1 d,7d,14d,and 28d after operation,respectively.The tissue bo-mogenate concentration of iNOS in the abdominal aortic aneurysm were detected by enzyme linked immu-nosorbent assay (ELISA).Results In experimental group,mean concentration of the tissue bomogenate concentration of iNOS in the abdominal aortic aneurysm in 1 d,7 d,14 d,28 d were (22.129 ±2.518)μ/mL,(27.337±5.321) μ/mL,(36.047±4.584)μ/mL,(44.756±1.799)μ/mL,respectively;In control group,that was (12.499±1.807)μ/mL.The concentration of iNOS in experimental group was significantly higher than that of control group (P < 0.01).The difference during the experimental group all had statisti-cal significance (P < 0.05).Conclusion It is possible that iNOS has some biological function during the formation and progression of the abdominal aortic aneurysm.

ABSTRACT

Objective To study the effect of noise on the ability of learning and memory of mice,and observe the effect of herba Houttuyniae injection on the change of the ability of learning and memory in noise condition, and then analyses the mechanisms. Methods To choose suitable mice and divide into groups, applying Y-maze and colorimeter to determine the content of MDA and the enzyme activity of SOD, AChE and NOS of mice brain. Results Learning group in Y-maze, noise cause the ability of noise group (12.1±1.20) decrease(P<0.01) compared with the control group(15.1±1.45), but herba Houttuyniae injection can reverse the ability of the mice, the correct frequency is much higher(P<0.01)in herba Houttuyniae group(14.9±1.60). The results is same in the memory group, in Y-maze, noise cause the memory ability of noise group (5.20±1.40) decrease(P<0.01) compared with the control group(6.30±1.32), but herba Houttuyniae injection can reverse the memory ability of the mice, the correct frequency is much higher(P<0.01)in herba Houttuyniae group(6.10±1.10). Either learning or memory group mice, noise can decreased the activity of SOD and NOS of mice brain(P<0.01), but the content of MDA and the ability of AChE elevated(P<0.01). herba Houttuyniae injection can change the effect of noise to mice, the activity of SOD and NOS of mice brain elevated(P<0.01) and the content of MDA and the ability of AChE decreased (P<0.01). Conclusion herba Houttuyniae injection can prevent the ability of learning and memory of mice which decreased by noise, and have the effect of intoxication.

@#Objective To observe the expression of induced nitricoxide synthase(iNOS)in ligustrazine against cisplatin-induced ototoxicity.Methods Fifty white guinea pigs with normal hearing were randomly divided into the control group,ligustrazine group and traditional Chinese drug(TCD)group.The alternations of hearing function were evaluated by the examination of auditory brainstem response(ABR).Some specimens were perfused by AgNO3 and observed the damage of bristle cell.And some were sent for immunohistochemistry to detect the expression of iNOS.Results Compared with the control group,the threshold of ABR in ligustrazine group and TCD group were significantly different.There was a significant difference between the ligustrazine group and TCD group.There was no expression of iNOS in Cortis',spiral ganglion cell and stria vascularis in normal cochlear(control group).In the ligustrazine group,the expression became stronger in spiral ganglion cell,stria vascularis,spiral ligament and Cortis'.INOS also expressed in TCD group.Conclusion Ligustrazine can antagonize cisplatin-induced ototoxicity by inhibiting the expression of iNOS.

Both carvedilol and metoprolol exhibited no effect on eNOS activity in all the investigated cardiac myocytes. CONCLUSIONS: Nebivolol does no effect on eNOS activity of left ventricular cadiocytes in subjects or rats without hear failure but it can inhibit eNOS activity of cadiocytes in subjects or rats with heart failure so as to exert its beneficial clinical effect.

ABSTRACT

Objective To observe whether piperine has the ability to prevent delayed cerebral vasospasm(DCVS) after subarachnoid hemorrhage(SAH).Methods Thirty-two New Zealand white rabbits were divided into four groups randomly(n=8).Group A(shamed-injecting blood): Puncture was completed without injecting blood into the cisterna magna;Group B(SAH): DCVS model was made with two-hemorrhage of SAH by injecting autologous arterial blood into the cisterna magna;Group C(SAH + piperine): Piperine(piperine(20 mg/kg),solvent(0.5 mL/kg)) was intravenously administrated daily since day 0 after first injection.Group D(SAH+DMSO): The solvent((0.5 mL/kg)) was gave by i.v.for DCVS models.All animals were sacrificed at day 7 after the first injection.The internal diameter and D/T(internal diameter/wall thickness) values of basilar artery(BA) were measured.The expression of ET-1 and eNOS in arterial wall of BA at day 7 was collected via immunohistochemistry method.Results The severe vasospasm of BA,over expression of ET-1 and diminished expression of eNOS in group B were found vs group A.The vasospasm of BA in group C was obviously alleviated in comparison with it in group B and D,respectively(P

ABSTRACT

PURPOSE: As suggested by many investigators, the cause of age-related female sexual dysfunction is the decline of serum estrogen. We investigated the effects of estrogen on the nitric oxide synthase (NOS) and vascular endothelial growth factor (VEGF) expressions and the histologic composition in the uterus and proximal and distal vagina of rats. MATERIALS AND METHODS: Forty-five mature female Sprague-Dawley rats were randomly divided into three groups; the control group (C), the oophorectomy group (O), and the estrogen replacement group (E). The expression rates of neuronal NOS (nNOS), endothelial NOS (eNOS), VEGF and the changes of the collagen component in the uterus and proximal and distal vaginal tissues were investigated by immunohistochemical staining and Masson's trichrom staining. RESULTS: The expression rate of NOS was highest in the proximal vagina, and this was followed by the portion of the vagina distal to the uterus. The nNOS and eNOS expression rates were significantly decreased in the uterus and the proximal vagina of the E group (p<0.05), and they were increased in the uterus and proximal vagina of the O group (p<0.05). However, the VEGF expression rate was increased in only the uterus of E group. In all tissues, the collagen component was significantly decreased in the E group, while it was increased in the O group (p<0.05). CONCLUSIONS: These data suggest that estrogen downregulates nNOS and eNOS. In addition, estrogen deficiency induces collagen synthesis and it decreases the content of smooth muscle in the uterus and vagina, resulting in structural relaxation difficulty.

Aim To observe the protective effect of valsartan,benazepril and combination valsartan withbenazepril on cardiac damage in rats with chronic renal failure and explore the mechanism.Methods Forty Sprague Dawley male rats were selected,and were performed five-sixths nephrectomy to produce chronic renal failure model.Two weeks after the surgery,the rats were divided randomly into model group,valsartan group,benazepril group and both valsartan and benazepril group,and a sham group was established as control group.Systolic blood pressure,BUN and serum creatinine was measured at 10 week after operation,then all rats were killed to take the hearts for pathological histological observation.The transcription of endothelin-1(ET-1 )mRNA and nitricoxide synthase (eNOS-3) mRNA in myocardium was examined by hybridization in situ.Results Systolic pressure,heart weight,heart weight index,left ventricular weight mass(LVWM) and left ventricular weight mass index(LVWMI) in model group increased significantly at 10 week after operation. AIM:To observe the neuroprotection effects exogenous adrenomedullin(AM) against hypoxic-ischemia reperfusion damage(HIRBD) in neonatal rat models.METHODS:Forty neonatal rats were randomly divided into Control Group(n=8,with sham operation),Hypoxic-ischemia Reperfusion(HIR) Group and Intervention Group(n=8,wth intraperitoneal AM injection 0,6 and 12 h after making model).All rats were decapitated at 48 hours after reperfusion,and the cerebral slices were made at the 2 mm level in front of the interaural line.The pathobiological changes of brain tissue were observed by the method of HE dye in light microscope,and the levels of nitrogen monoxide synthase(NOS),glutathion(GSH) and myeloperoxidase(MPO) in neonatal rats cerebral tissue were determined by using chromatometry.RESULTS:Hypoxic-ischemia reperfusion could induce obviously cerebral pathological change which could be improved by AM.The brain tissue levels of cNOS and GSH in AM 0 and 6 h Group were significantly higher than those of HIR Group(P

ABSTRACT

Objective To study the protective effect of total flavone from Rhododendron simsii(TFRS) on myocardial ischemia-reperfusion injury rats and its mechanism.Methods The ischemic model was made by occluding the anterior descending of the left artery(LAD) in rats.The change of ST segment and T wave of electrocardiograph(ECG) were observed,and the activity of lactate dehydrogenase(LDH),creatine kinase(CK),levels of the maleic dialdehyde(MDA),and nitric oxide(NO) in serum were measured.And by tetrazolium chloride(TTC) staining,the areas of myocardial infarction were observed.The expression of inducible nitricoxide synthase(iNOS) in rats was detected by emploring the reverse transcription-polymerase chain reaction(RT-PCR) technique.Results On the myocardial infarction model by occluding the anterior descending of the LAD in rats,TFRS(100 mg/kg) obviously reduced the height of ST segment after occluding 30 min and TFRS(25,50,and 100 mg/kg) obviously reduced the height of ST segment after reperfusion 30 min.TFRS(50 mg/kg) reduced by myocardial infarction area.TFRS(50 and 100 mg/kg) obviously reduced the activity of CK and LDH.TFRS(50 mg/kg) decreased the level of MDA in serum.By RT-PCR technique,it was found that the expression of iNOS mRNA in myocardium in IR rats pretreated with TFRS(100 mg/kg) was higher than that in IR and normal groups.Conclusions TFRS has the significant protection against myocardial ischemia-reperfusion injury via atte-nuating oxygen free radical and increasing the expression of iNOS mRNA and NO production.

ABSTRACT

Objective To investigate the protective effects of felodipine on mRNA levels of endothelial nitricoxide synthase (eNOS) and inducible nitricoxide synthase (iNOS) as well as the level of nitrogen monoxidum (NO) from human umbilical vein endothelial cells (HUVECs) injuryed by oxidized low density lipoprotein (ox-LDL). Methods Isolated HUVECs were divided into blank control group,ox-LDL injury group treated with ox-LDL of different concentrations (6,12.5 and 25 mg/L),and intervention group of felodipine (0.1,1.0 and 10 ?mol/L)+ox-LDL (25 mg/L). Then eNOS and iNOS expressions were measured by real time-polymerase chain reaction and the level of NO in the supernatants of the cultures was assayed by nitrate reductase method. Results The mRNA expressions of eNOS and iNOS in HUVECs and NO level in the supernatants during treatment with different ox-LDL concentrations were higher than those in control group. However,felodipine significantly down-regulated the expression of iNOS in HUVECs injured by ox-LDL and inceased NO generation. Conclusion Felodipine has protective effects on endothelial cells. The mechanism may be related to its lowering the mRNA expression of iNOS induced by low ox-LDL concentration and increasing NO production.
